赖氨酸甲基转移酶SET8结构和功能及其与肿瘤关系的研究进展

Association of Structure and Functions of Lysine Methyltransferase SET8 with Tumorigenesis

  • 摘要: SET8(又称SETD8、PR-SET7、KMT5a)是现今发现唯一能够特异性单甲基化H4K20的赖氨酸甲基转移酶。SET8及其催化形成的H4K20me1共同参与了细胞周期调控、染色质结构及基因转录的调节。近年来研究发现SET8在多种肿瘤组织中高表达,参与调控体内细胞的细胞周期、增殖及凋亡等过程,涉及肿瘤的发生、生长、转移等多个环节,有望成为肿瘤治疗的新靶点。本文主要从SET8的结构、功能及其在肿瘤发生中的作用等方面综述了SET8的研究进展。

     

    Abstract: SET8, also named SETD8, PR-SET7 or KMT5a, is a sole lysine methyltransferase that catalyzesmonomethylation of histone H4 lysine 20(H4K20). SET8 and the H4K20me1 catalyzed by SET8 are jointly implicated in modulating cell cycle, maintaining chromatin structure and regulating the transcription of several genes. Recently, it is reported that the high expression of SET8 involves in regulating cell cycle, cell proliferation and apoptosis. SET8 is also related with tumorigenesis, progression and metastasis, suggesting that SET8 may be a potential target for cancer treatment. In this review, we focus on analyzing the association of the structure and function of SET8 and its effect in tumorigenesis.
