膀胱癌肿瘤干细胞筛选方法的研究进展

Recent Progress on Isolating Bladder Cancer Stem Cells

  • 摘要: 膀胱尿路上皮癌(bladder cancer, BCa)是最常见的泌尿系统恶性肿瘤,BCa术后容易复发、转移的原因可能是BCa的“肿瘤干细胞”(cancer stem cells, CSCs)没有被清除或消灭。在研究膀胱癌肿瘤干细胞(BCa-CSCs)的过程中,如何筛选得到BCa-CSCs是非常重要的。本文就目前BCa-CSCs筛选方法的研究进展进行总结,包括细胞表面的特异标志物、侧群细胞分选法、无血清悬浮培养法、化疗药物法和其他可以借鉴的筛选方法,如吸附能力梯度法、侵袭能力差异法、Fibronectin差别黏附法等。目前这些方法都有不足之处,需不断摸索、完善、优化,从而得到一种最佳的Ba-CSCs的筛选方法。

     

    Abstract: Bladder cancer (BCa) is the common malignant urologic cancer in the world. The reason for easy relapse and metastasis of BCa after surgery could be a failure to eliminate the cancer stem cells (CSCs). How to isolate BCa-CSCs is rather important during investigation. This review summarizes the recent progress of BCa-CSC isolation methods, including stem cell surface markers, side population cells identification, the serum-free medium suspension culture, chemotherapeutics, and other methods which could be referred such as adsorption capacity gradient, in invasion ability differential, fibronectin differential adhesion assay and so on. Currently owing to the shortcomings of these methods, it would be reasonable to learn from other isolating methods to obtain an optimal one for Ba-CSCs isolation.
