代谢综合征和乳腺癌之间的因果关系:双向双样本孟德尔随机化

Causal Relationship between Metabolic Syndrome and Breast Cancer: A Bidirectional Two-Sample Mendelian Randomization

  • 摘要: 目的 分析代谢综合征与乳腺癌二者间是否存在因果关系。方法 分别从IEU GWAS数据库和GWAS catalog数据库下载代谢综合征、乳腺癌的GWAS数据,采用双向设计,二者互为因果,互相作为暴露与结局数据来进行分析,同时采用逆方差加权法为主, MREGGER法、加权中位数法、简单模式法和加权模式法为补充,对F>10 的强工具变量与乳腺癌进行双向两样本孟德尔随机化分析, 并且通过异质性分析、水平多效性分析、森林图和逐一剔除验证方法来分析其结果的可靠性。结果 IVW分析显示,代谢综合征与乳腺癌之间并不存在因果关系 OR=1.00,95%CI(0.97-1.03),P>0.05。敏感性分析显示结果可靠。结论 本研究未发现证据支持代谢综合征与乳腺癌之间存在正向因果关系,也未能支持乳腺癌与代谢综合征之间存在反向因果关系。

     

    Abstract: Objective: To investigate the causal relationship between metabolic syndrome and breast cancer. Methods: GWAS data for metabolic syndrome and breast cancer were downloaded from the IEU GWAS database and the GWAS catalog database, respectively. A bidirectional design was employed, treating each as both exposure and outcome. Two-sample Mendelian randomization analysis was performed, using inverse variance weighted method as the primary approach, supplemented by MR-Egger regression, weighted median, simple mode, and weighted mode methods. Strong instrumental variables (F>10) were analyzed. Heterogeneity analysis, horizontal pleiotropy analysis, forest plots, and leave-one-out validation were used to assess the reliability of the results. Results: IVW analysis showed no causal relationship between metabolic syndrome and breast cancer OR=1.00, 95%CI (0.97-1.03), P>0.05. Sensitivity analyses confirmed the robustness of the findings. Conclusion: This study found no evidence to support a positive causal relationship between metabolic syndrome and breast cancer, nor did it support a reverse causal relationship between breast cancer and metabolic syndrome.
