基于NF-κB信号通路探讨中药治疗肝癌的研究进展

Research Progress on the Treatment of Liver Cancer with Mechanistic Insights into Herbal Medicine for Hepatocellular Carcinoma: Focus on the NF-κB Signaling Pathway

  • 摘要: 肝癌是全球范围内发病率和死亡率较高的恶性肿瘤之一,其治疗面临巨大挑战。NF-κB信号通路在肝癌的发生、发展及治疗中扮演重要角色,其异常活化与与肿瘤细胞增殖和凋亡有着紧密的关联。近年来,研究表明通过NF-κB信号通路抑制肝癌有一定成果。而中药因其多种类、多效用的作用特点,在肝癌治疗中展现出独特优势。本文综述了NF-κB信号通路在肝癌中的作用机制,以及中药通过调控NF-κB信号通路治疗肝癌的研究进展。研究表明,中药通过抑制NF-κB信号通路的活化,诱导肝癌细胞凋亡,从而发挥抗肿瘤作用。此外,中药还通过与其他信号通路的交互作用,增强抗肿瘤效果。本文旨在为中药单体及复方抗肝癌研究提供理论依据和新思路,推动中药现代化与国际化进程。

     

    Abstract: Hepatocellular carcinoma (HCC) ranks among malignancies with high global incidence and mortality rates, presenting significant therapeutic challenges. The NF-κB signaling pathway plays a pivotal role in HCC pathogenesis, progression, and treatment resistance, where its aberrant activation critically correlates with tumor cell proliferation and apoptotic dysregulation. Recent studies have demonstrated promising therapeutic outcomes through NF-κB pathway inhibition in HCC. Meanwhile, Traditional Chinese Medicine (TCM), leveraging its multi-component nature and multi-target effects, exhibits unique advantages in HCC management. This review synthesizes the mechanistic roles of the NF-κB pathway in HCC and current advances in TCM-mediated HCC treatment via NF-κB modulation. Evidence confirms that TCM agents exert antitumor effects by suppressing NF-κB activation and inducing HCC cell apoptosis, while further enhancing efficacy through crosstalk with complementary signaling pathways. This work aims to provide theoretical foundations and novel perspectives for developing TCM-derived active monomers and compound prescriptions against HCC, thereby advancing the modernization and global integration of TCM therapeutics.
