Research on the Induction of Apoptosis by the Decoction of Rhizoma Zedoariae and Fructus Trichosanthis in PGLH7 cell by Serologic Pharmacology

Abstract
Objective To investigate the mechanism of the decoction of Rhizome Zedoariae and Fructus Trichosanthis in the induction of apoptosis of PGL H7 cell by serologic pharmacological method. Methods After incubating PGL H7 cell and Chinese drug, borne rabbit serum at different dose with serologic pharmacological method. The apoptosis morphological changes were observed under the inverted microscope, light microscope and fluorescence microscope in vit ro. The target cells were harvested to estimate the apoptosis ratio by flow cytometry. Results The PGL H7 cell showed the typical apoptosis morphological changes, such as shrinkage, crescent shape concent rated chromatin and fragmentation af ter being treated with drug-bearing serum with the inverted microscope and light microscope. Fluorescence microscope and staining with Annexin V & PI are used to observe the typically early and final phase changes of PGL H7 cell apoptosis. There was significant difference in cell apoptosis ratio between 10 % high, middle dose drug-bearing serum and the cont rol serum by flow cytomet ric assay. The effect was dependent on both time and dose. Conclusion The antineoplastic molecular mechanisms of the decoction of Rhizome Zedoariae and Fructus Trichosanthis may be its effect on inducing the apoptosis of PGL H7 cells by serologic pharmacological method.
