Expressions of DR5,caspase-8,caspase-9 and caspase-3 in HepG2 Cell Line Treated with Gemcitabine

  • ObjectiveTo research DR5,caspase-8,caspase-9 and caspase-3 expressions in HepG2 cells treated by gemcitabine,and preliminary apoptosis mechanism induced by gemcitabine. Methods After treated by virous cencetrations,HepG2 cell growth inhibition and apoptosis was assessed by MTT and FMC respectively;the expressions of DR5,caspase-8,caspase-9 and caspase-3 were detected by immunohistochemical method and Western blot. Results Gemcitabine apparently inhibited the growth of HepG2 and induced apoptosis with a time-dependent and concentration-dependent way After treated by gemcitabine for 24 h,the expressions of DR5,caspase-8,caspase-9 and caspase-3 were up-regulated. Conclusion Gemcitabine induced the apoptosis of HepG2 through up-relation of DR5,caspase-8,caspase-9 and caspase-3.
