Effect of hTERTC27 Overexpression on Prolifteration and Apoptosis of Nasopharyngeal Carcinoma C666-1 Cell Line

Abstract
Objective To explore the effect of hTERTC27 overexpression on the proliferation and apoptosis of nasopharyngeal carcinoma C666-1 cell line. Methods pcDNA3-hTERTC27 was transfected into nasopharyngeal carcinoma C666-1 cells with lipofectamine.The expression of hTERTC27 and Bcl-2 were detected by Western blot.The proliferation of cells was detected by MTT assay and colony formation assay.The apoptosis of cells was detected by TUNEL assay. Results C666-1 cells expressed high level of hTERTC27 after 24h transfection.Over expression of hTERTC27 obviously suppressed the cell proliferation(P<0.01),downregulated the colony formation rate(P<0.01),increased the TUNEL positive cells (P<0.01),and downregulated the expression level of Bcl-2 protein(P<0.01). Conclusion Overexpression of hTERTC27 can effectively inhibit proliferation and promote apoptosis of nasopharyngeal carcinoma cells,which is possibly due to the down regulation of Bcl-2 protein.
