Effect of Id1 on Proliferation and VEGF Expression of Colon Cancer HT-29 Cells

  • Objective To investigate the role of inhibitor of differentiation-1 (Id1) on proliferation of colon cancer HT-29 cells,and detect the effect of Id1 on expression of VEGF in HT-29 cells. MethodsThe expressions of VEGF mRNA and protein were detected by RT- PCR and Western blot assay in HT-29 cells after transfection with Id1 expression vector or Id1-siRNA.The impact on proliferation of HT-29 cells was investigated by MTT assay. Results The overexpresssion of Id1 promoted both the mRNA and protein of VEGF and Id1 suppression significantly reduced both the mRNA and protein level of VEGF.MTT assay showed that the cell proliferation of suppression group was significantly slower than that of control group. Conclusion Id1plays an important role in colon cancer proliferation by up-regulate the expression of VEGF in HT-29 cells.Id1 is expected to become a new target for colon cancer therapy.
