Nude Mice Transgenic Hepatic Metastasis Model of Colon Cancer

  • Objective To explore the method to establish transgenic animal model for liver metastasis of colon cancer. Methods After establishing transgenic models for liver metastasis of human colon cancer Lovo cell which transfeced with CD gene in nude mice in vitro and in vivo, the rate of liver metastasis and the life span were detected between two groups. Pathological features were observed by HE stain. The RNA and protein expression of CD gene was analyzed by Real-time quantitative RT-PCR and Western-blot. Results No significant difference were observed between both groups about the rate of liver metastasis and the life span 100%(15/15),100%(15/15); (40.60±2.2297)d, (41.73±2.4919)d; P>0.05)]. The RNA expression of CD gene in vivo was lower than that in vitro (P<0.05,t=2.758). The protein expression level of CD gene in the tumor tissue detected by westernblot was lower in vivo than that in vitro. Conclusion CD gene can more stably and persistently express in the Lovo cell transfected in vitro than that transfected in vivo.
