Effect of Docetaxel Combined with Mitomycin C on Proliferation and Migration of HeLa Cells

Abstract
Objective To investigate effect of Docetaxel (Doc) combined with Mitomycin C (MMC) on pro2 liferation and migration of HeLa cells. Methods Proliferation and migration ability of HeLa cells were tested by MTT and wound healing assay respectively. The combined effect of Doc and MMC was evalua2 ted by“Q value”. Results The inhibition effect of Doc or MMC on proliferation of HeLa cells was dose dependent (Doc :0. 5μg/ L vs 0. 1μg/ L, P < 0. 05 ;MMC:10μg/ L vs 2. 5μg/ L, P < 0. 05) ;“Q value”ex2 ceed 1. 0, which indicated the synergistic effect of Doc and MMC. The migration velocity of cells in (Doc + MMC) group (0. 035μm/ h) was lower than Cont rol (0. 173μm/ h, P < 0. 05) group, Doc (0. 079μm/h) and MMC (0. 075μm/ h) group. Conclusion Doc and MMC dose2dependently inhibit proliferation and migration of HeLa cells respectively, and there was synergistic effect of the combination therapy. That may be on account of the different target s of two drugs.
