胃癌单克隆抗体MG_cl相应抗原的初步研究

摘要: 本文报告一株鼠抗人胃癌单克隆抗体MG_cl相应抗原的纯化及分析。MG_cl经50%饱和硫酸铵盐析，DEAE—52纤维素柱层析后，获得纯化的IgG56，31mg与2g溴化氰活化的sepharose 4B偶联，制备免疫吸附剂。用MG_cl免疫亲和层析的方法从胃癌组织中纯化出MG_cl相应抗原，经蛋白酶处理，高碘酸氧化，电泳脂蛋白袭色，SDS—PAGE及Immunoblotting等实验分析证实MG_cl相应抗原是一种低分子量的糖蛋白，分子量为72Kd和52kd。

Abstract: The corresponding antigen of a monoclonal antibody (MGcl)against gastric cancer was purified and partally characterized. The MGcl was purified by passing throuth a DEAE cellulose column and covalently coupled to CNBr activated Sepharose 4B. By means of immunoaffinity chromatogrphy,the Corr- esponding antigen of MGcl was purified from gastric cancer tissue. The results of periodate oxidation,SDS-PAGEand Immunblotting studies suggest that the corresponding antigen of MGcl is a glycoprotein with molecular weight of 72 KD and 52 KD.
