结肠癌细胞株、癌组织及正常结肠粘膜中P16基因异常甲基化率的差异

The Difference of P16 Gene' s Aberrant Methylation Rate Between Colon Cancer and Normal Colonic Mucosa

  • 摘要: 本文采用Southernblot法鉴定了结肠癌细胞株、癌组织及正常结肠粘膜组织中P16基因的甲基化情况。结果在6株结肠癌细胞株中有5株(83.3%)P16基因CpG岛呈异常甲基化状态,在18例结肠癌组织中13例(72.2%)呈异常甲基化状态。与之相对,18例正常结肠粘膜组织中仅5例(27.8%)表现为异常甲基化,提示在结肠癌中P16基因因异常甲基化而失活,这为结肠癌的基因治疗提供了新策略、新目标。

     

    Abstract: The methylation rate of P16 gene in colon cancer cell lines, colonic cancer tissue and norm al colonic mucosa were examined by Southern blot method. T he data showed that aberrant methylation of 5' CpG island of P16 gene was observed in5 of 6(83.3%)colon cancer cell lines, 13 of 18(72. 2%)colon cancer tissue.In contrast,only 5 of 18(27.8%)normal colonic mucosa showed aberrant methylation of the 5'CpG island of P16 gene.The results suggest that P16 gene in colon cancer was inactivated by aberrant methylation.This can provide a new tactics and a new target for colon's gene therapy.
