Significance of HLA-DR, Fas, bcl-2 and bax Expression in Hepatocellular Carcionma

  • 摘要: 目的:探讨人白细胞抗原(HLADR抗原)和凋亡相关基因Fas、bcl2及bax在肝癌中表达的意义。方法:应用免疫组化方法研究40例肝癌组织中HLADR、Fas、bcl2及bax的表达。结果:40例肝细胞癌中18例HLADR阳性,阳性率45%,癌旁肝细胞阴性。6例癌细胞Fas阳性,阳性率为15%,癌旁细胞20例阳性,阳性率500%。癌细胞bcl2阳性6例,阳性率为15%,8例癌旁细胞阳性,阳性率为20%。癌细胞bax阳性8例,阳性率为20%,癌旁细胞25例阳性,阳性率为625%。结论:提示HLADR异常表达与肝细胞癌变有关;Fas可能不是引起肝癌细胞凋亡的主要途径;bcl2和bax的变化可能影响肝细胞的凋亡状态,在肝癌发生中可能起一定作用。

     

    Abstract: In order to investigate the significance of HLA-DR, Fas, bd-2 and bax expression in hepatocellular carcinoma (HCC).Immunohistochemical analysis with monoclonal or polyclonal antibodies for HLA DR, Fas, bd-2 and bax were used to detected the protein in 40 cases of HCC .The results showed that 18 cases in tumor cells were positive for H LA-DR antigen (positive rate 45%),but no expression in adjacent tissues. 6 cases in tumor cells and 20 cases in adjacent tissues were positive for Fas(positive rate 15%and 58.8%)6 cases in tumor cells(15%)and 8 cases in adjacent tissues(20%)were positive for bcl-2.8 cases in tumor cells (20%)and 22 cases in adjacent tissues (62.5%)were positive for bax.results indicated that(1).Aberrant expression of HLA-DR antigen may be related to tumorigen esis of HCC.(2)Fas may not play amajor role in apoptosis in HCC.(3)The changes of bcl-2 and bax expression may play a role in the tumorigeresis of HCC by effecting on the apoptosis state of hepatocyte.
