5-氟尿嘧啶诱导人卵巢癌细胞凋亡的实验研究

Apoptosis in Human Ovarian Carcinoma Cells Induced by 5━fluorouracil

  • 摘要: 用不同剂量的5-氟尿嘧啶(5—Fu)处理人卵巢癌AO10/17细胞株,观察药物作用后细胞形态学,DNA片段化和细胞内钙离子浓度的变化。结果表明受药物影响的人卵巢癌细胞内ca++升高,出现了细胞凋亡的形态改变。最后探讨了5—Fu抑制卵巢肿瘤细胞的作用机理。

     

    Abstract: The induction of apoptosis in human ovarian carcinoma cell line AO10/17 was morphologically and biologically examined by 5-fluorouracil (5—Fu).It has shown that AO10/17Underwent apoptosis as characterized by morphologic changes with chromatin condensation and DNA fragmetation after treated by 5-Fu.A flow cytometric method formeasuring the percentage of apoptotic nuclei (hypodiploid DNA)after PI staining and colormetric method measure DNA fragmentation in the nucleatr extracts,both of them were higher in treated cells than is untreated cells.The date demonstrates effection of 5—Fu on AO10/17 is associated with apoptosis.
