Obeservation of Phenotypic Changes of Human Hepatocarcinoma Cell Line Induced by Sodium Butyrate

  • 摘要: 目的 探讨SB对人肝癌细胞的诱导分化作用。方法 以0-2mmolL的SB作用于培养的人肝癌细胞GHC-3,作用30 天后恢复正常培养,观察癌细胞表型的变化。结果 人肝癌细胞生长抑制;SB作用30 天细胞形态由多角型转变为棱形,ConA凝集现象消失,琼脂集落形成率下降;染色体主流范围变窄,二倍体及亚二倍体比例升高;3H-TdR 掺入率、细胞DNA含量下降与对照比较有显着差异( P< 0-01) ;并观察到,上述细胞再经不含SB 的正常培液继续培养三代后该细胞表型未发生明显改变。结论 人肝癌细胞在低剂量丁酸钠持续作用下,某些恶性表型可发生部分逆转,且这些改变在恢复正常培养后可保持相对稳定。

     

    Abstract: Objective Dectect the Effect of sodium butyrate(SB) on the phenotypes in human hepatocarcinoma cell line(GHC-3). Methods The cells were treated by 0 2mmo/L sodium butyrate for thirty days and then transferred back to the standard medium for fifteen days. Results After GHC-3 cells were treated by SB thirty days,Morphology changes from polygonal to spindle-shaped,proliferation was inhibited significantly,colony-forming rate decreased,ConA agglutinability disappeared,the major range of chromosome turned from 52~59 to 44~45,the pencentages of diploid and hypodiploid increased;the 3 H-TdR incorporation and DNA concentration decreased markedly.These induced phenotypes did not change after transferred back to the standard medium for fifteen days. Conclusion The malignant phnetypes of the human hepatocarcinoma cell line,GHC-3,can be reversed partly and these reversed phenotypes can keep relativityly stability.
