香烟对阻止黄曲霉素B1诱致小鼠肝脏癌前病变影响的bFGF表达

Effect of Tobacco Smoke on Basic Fibroblast Growth Expression of AFB1 Induced Precancer Cells in Mouse Liver

  • 摘要: 目的:应用抗bFGF单克隆抗体研究小鼠肝脏癌前病变中bFGF的表达。方法:将津白2纯系小白鼠60只随机分成三组:AFB1组,AFB1+香烟熏组和对照组。AFB1溶于DMSO,5ug/日,只量拌于饲料投食两实验组,熏烟组另以烟丝30g/日熏烟,经半年实验,取肝脏切片行免疫组化染色。使用医学图像处理系统检测bFGF表达强度。结果:三组小鼠肝bFGF表达强度呈AFB1组>熏烟组>对照组的趋势,各组间差异均具有显著性意义(P<0.05),且bFGF表达与DNA量呈正相关。我们推测:AFB1诱导的小鼠肝脏癌前病变与bFGF表达上调所至细胞增殖有关。结论:本实验从另一侧面证实香烟具有阻止AFB1诱导小白鼠肝癌变的作用。

     

    Abstract: In this study,60 pure bred mice were divided into three groups 20 each:(1) AFB1 infected group,(2) AFB1 plus tobacco smoking group and (3) Control group.Crystalline AFB1 was dissolved in DMSO and fed the experimental mice.An affinity purifed polyclonal rabbit antiserum to human bFGF was used as primary antibody with immunohistochecal staining,bFGF was frequently detected in the cytoplasm of AFB11 infected group.The expression intensities were shown as follows:AFB1 infected groupcontrol and AFB1 plus tobacco smoking group.The results showed the expression of bFGF demonstracted a close correlation to the degree of malignancy.The bFGF expression in each group showed positive correlation with nuclear DNAcontent.So we proposed that cigarette smoking might be protected against AFB1 induced primary cancer of the liver in mice.
