VEGF反义核酸与柔红霉素联用对增加髓性白血病细胞化疗敏感性的研究

Study of VEGF Antisense Oligodeoxynudeotide and Daunorubicin to Increase the Sensitivity of AML and CML Cells

  • 摘要: 目的 探讨血管内皮生长因子VEGF(VascularEndothelialGrowthFactor)反义核酸与柔红霉素联用, 对AML、CML细胞药物敏感性的影响。方法 采用以前实验筛选所得的最优反义核酸A7,长度为 2 0脱氧核糖核苷酸, 全硫代修饰 ;以脂质体介导转染细胞, 2 4h以后加入柔红霉素, 继续培养 72h, 用0 .4 %苔盼蓝拒染法计数活细胞, 用ELISA法检测培养液中VEGF蛋白的浓度, 用流式细胞仪检测细胞凋亡百分数。结果 VEGF反义核酸显著增加柔红霉素抑制AML、CML细胞生长, 下调VEGF蛋白的表达, 诱导AML、CML细胞凋亡。结论 VEGF反义核酸提高AML、CML细胞对柔红霉素敏感性 ;内源性VEGF蛋白使AML、CML细胞产生耐药性

     

    Abstract: Objective To explore the effect of VEGF antisense on sensitivity of AML and CML cells to Daunorubicin (DNR).
