无血清培养基与完全培养基体外诱导扩增CIK细胞分泌细胞因子水平的比较

Comparison of Serum-Free Medium AIMV with Standard Serum-Containing Medium in Proliferation and Excretion Cytokine Level of CIK Cells in Vitro

  • 摘要: 目的比较无血清培养基(AIMV)与完全培养基(CM)体外诱导扩增CIK细胞及CIK细胞分泌细胞因子水平。方法分别用AIMV及完全培养基加入4种细胞因子(IFN-γ、IL-2、IL-1及OKT3)将脐带血单个核细胞(CBMNCs)诱导成CIK细胞,比较细胞的增殖能力、细胞表型及分泌细胞因子水平。结果与完全培养基比较,无血清培养基培养的CIK细胞增殖高峰较晚(14~17天),但增殖倍数高,在细胞表型、分泌细胞因子水平方面两种培养基培养的CIK细胞均无明显的差别(P>0.05)。结论无血清培养基可代替完全培养基。

     

    Abstract: Objective To compare serum-free medium AIMV with standard serum-containing medium in proliferation CIK cells and excretion cytokine level in vitro. Methods CIK cells were generated by culture CBMNCs in the presence of four cytokines(IFN-γ,IL-2,IL-1 and OKT3) in serum-free medium AIMV or in standard serum-containing medium separately.
