Effect of PDGF on the in-Vitro Growth of Leukemic Cell Dami

  • 摘要: 目的 本文试图了解PDGF对肿瘤细胞体外生长和胞内传递通路的作用及肿瘤细胞是否表达PDGF受体。方法 应用白血病细胞株Dami作为肿瘤细胞模型, 观察PDGF对Dami细胞的体外增殖和集落生成的影响, 并应用Westernblot方法分析对胞内c fos蛋白的作用。亦使用流式细胞术和免疫组化方法分析PDGF受体的表达。结果 Dami细胞表达PDGF受体, 而且PDGFβ受体水平高于PDGFα受体水平。通过该受体, PDGF上调胞内c fos蛋白的表达和促进Dami细胞增殖和集落生成。结论 PDGF可能通过肿瘤细胞表面的PDGF受体, 激活胞内信息通路, 刺激细胞的增殖和集落形成。提示血小板在肿瘤的发生发展中有一定的作用

     

    Abstract: Objective It could help us to understand the role of PDGF in leukemogesis. Methods Dami cells were used as a tumor cell model to investigate the effect of PDGF on this cell proliferation and colony formation. Western blot was used to detect the effect of PDGF on c fos expression of these cells. Flow cytometry and immunocytochemical staining were also used to identify PDGF receptor in Dami cells.
