透明质酸酶对人乳腺癌细胞MDA231增殖和转移的影响

An experimental study of human membrane-bound hyaluronidase (PH-20) on breast cancer cell (MDA231) growth and metastasis

  • 摘要: 目的 观察透明质酸酶 (PH 2 0 )基因对人乳腺癌细胞在鸡胚胎中生长和转移的影响。方法 PH 2 0cDNA转染到人乳腺癌细胞株MDA2 31中 (MDA2 31 PH2 0 ), 细胞种植到鸡胚绒毛尿囊膜 (CAM )上, 观察肿瘤大小 ;对肿瘤组织血管内皮细胞染色, 研究肿瘤新生血管生长变化 ;PCR扩增远端CAM中AluDNA序列, 观察PH - 2 0对肿瘤细胞转移的影响。结果 MDA2 31 PH2 0在CAM上增殖的程度明显高于对照组, 血管内皮细胞数量也增多, 同时远端CAM出现AluDNA序列, 而对照组无。结论 PH 2 0明显促进癌组织内新生血管生长, 并促进乳癌细胞增殖和远端转移。

     

    Abstract: Objective To study the effects of human sperm membrane-bound hyaluronidase (PH-20) on gene transfected MDA231 cells growth and metastasis. Methods PH-20 cDNA was transfected into human breast cancer cell line MDA231.Transfectants (MDA231-PH-20) were then implanted onto chorioallantoic membrane (CAM). Meanwhile, empty vector pcDNA3 was transfected into MDA231 (MDA231-pcDNA3) as control.
