rBCG-IL-2对肿瘤生长抑制作用和免疫刺激作用的研究

Study on the Effect of rBCG-IL-2 on Anti-tumor Growth and Immunine Stimulation

  • 摘要: 目的 将表达IL-2重组BCG用于动物实验,观察其对肿瘤生长和机体免疫功能的影响。方法 采用基因工程技术和电转化技术,构建重组卡介苗即rBCG-IL-2和rBCG,并用瘤株重组BCG接种荷瘤小鼠,几周后观察肿瘤生长情况和检测巨噬细胞的杀伤活性。结果 rBCG-IL-2能表达分泌IL-2,其表达量为9.87ng/ml±4.56ng/ml,rBCG-IL-2与rBCG抑制肿瘤生长的作用明显优于对照组,并能提高小鼠巨噬细胞杀瘤率。结论 rBCG-IL-2能有效抑制肿瘤生长,较早刺激机体的免疫功能,提高巨噬细胞的杀瘤活性。

     

    Abstract: Objective To inquire into the effect of rBCG-IL-2 on tumor growth and immunity after it has been inoculateded in animal. Methods Gene engineering and electric transformation technologies were adopted to construction recombinant BCG,one of which was rBCG-IL-2 strain and the other was rBCG strain.Observe the growth of tumor and determine tumoricidal effect of macrophage after the tumor cells and recombined BCG have been inoculated in mouse in a few weeks. Results rBCG-IL-2 expressed and secreted IL-2 and the count of secreting was about 9.87ng/ml±4.56ng/ml.The anti-growth effects of tumor of rBCG-IL-2 and rBCG were obvious higher than the control. Conclusion rBCG-IL-2 can prevent tumor from growing effectually,active immunity of the host early and enhance cytotoxicity of macrophage obviously.
