MMP-2、MMP-9酶活性与人肝细胞癌生物学行为的研究

Study on Relationship between Activity of MMP-2(72KD),MMP-9(92KD) and Invasion Metastasis of Human Hepatocellular Carcinoma

  • 摘要: 目的 探讨MMP-2(72KD)、MMP-9(KD)的活性与肝细胞癌(HCC)浸润、转移的关系。方法 利用酶谱和计算机软件检测并分析了32例肝细胞癌组织中MMP-2、MMP-9活性。结果 肝细胞癌72KD的水平(28.89±7.28)高于癌旁组(19.41±5.52),P<0.05;肝癌组62KD+72KD(55.46±11.64)与癌旁组(28.06±7.72)比较时,P<0.05。同时发现62KD恶性组(26.57±9.3)明显高于癌旁组(8.65±1.98),P<0.05;转移组62KD(34.72+13.52)明显高于非转移组(18±41)62KD/62KD+72KD;恶性组(0.47)明显高于癌旁组(0.31),P<0.01;转移组(0.53)明显高于非转移组(0.41),P<0.01。另外,肝细胞癌中92KD的水平(27.95±11.93)明显高于癌旁组(7.46±2.53),P均<0.01;转移组(35.59±13.67)高于非转移组(20.31±10.18)P<0.01。结论 结果提示MMP-2、MMP-9的水平及活性与肝细胞癌的浸润和转移有关。

     

    Abstract: Objective To investigate lelationship between activity of MMP-2(72KD),MMP-9(92KD) and invasion,metastasis of human hepatocellular carcinomar(HCC). Methods Useing Zymography and computer software assisted analysis,we have measureed the activitive levels of MMP-2(72KD),MMP-9(92KD)in tissues from HCC. Results Mean activitive levels of 72KD(28.89±7.82)in HCC were higher than those in benign disease,P<0.05.There was difference(P<0.05). in 62KD+72KD of the tumorous tissues and the non-tumorous tissues.62KD(26.57±9.30)was markly higher in HCC (P<0.01).We also fund 62KD/62KD+72KD were apparently higher in HCC(0.47) and metastases (0.41),P<0.01 ,respectively.Meanwhile,levels of 92KD were higher in HCC tiessues (27.95±11.93)than in nontumorous tissues(7.46±2.53),P<0.01.levels of 92KD were higher in metastases than in non-matastasis carcinomas,P<0.01,Conclusion These results demonstrate that levels of MMP-2,MMP-9 can be candicates for novel markeres for HCC.The levels appear to reflect their potential and angoing activity of invasion and metastasis of HCC.
