大豆异黄酮对乳腺癌细胞TGFβ表达的影响

Expression of Transforming Growth Factor β in Human Breast Cancer Cells Induced by Soybean Isoflavones

  • 摘要: 目的 探讨大豆异黄酮作用后 BCap- 37人乳腺癌细胞 TGFβ和 TGFβ受体表达的改变。方法 以二羟异黄酮、三羟异黄酮处理 BCap- 37细胞, 采用 TGFβ拮抗试验、Western blot分析和原位杂交等实验方法, 检测大豆异黄酮作用后 BCap- 37人乳腺癌细胞 TGFβ1、TGFβ2 及受体表达的变化。结果  3× 1 0 -5mol/L三羟异黄酮作用后 Bcap- 37细胞 TGFβ1、TGFβ2 及受体表达增强, 而二羟异黄酮处理组在相同作用浓度下其表达无明显改变。结论 三羟异黄酮可促进 Bcap- 37细胞 TGFβ1、TGFβ2 及受体的表达。

     

    Abstract: Objective To investigate the chang of expression of TGF- and TGF- receptor in the human breast cancer cells Bcap-37 induced by soybean isoflavones. Methods TGFβ1、TGFβI mRNA and protein of Bcap-37 cells were determined with In situ hybridization and immunoblotting after cells were treated with daidein or genistein for 1~2 days.The expression of TGF-β1 and TGF-β2 was determined with TGF-β resistance test.
