负载酸洗抗原肽的树突状细胞诱导抗膀胱癌的作用

Cytotoxicity Induced by Dendritic Cells Pulsed with Acid Eluted Tumor Antigen Peptides against Bladder Cancer Cell Line

  • 摘要: 目的研究弱酸洗脱法提取抗原肽致敏的树突状细胞(DCs)诱导针对膀胱癌细胞的特异性杀伤效应。方法弱酸洗脱法获得BI U-87细胞抗原肽,用GM-CSF、IL-4和TNF-α体外诱导正常人DC并负载肿瘤抗原,进而激活T淋巴细胞;用MTT法检测其对BI U-87体外杀伤效应。结果弱酸可以洗脱肿瘤细胞膜表面大部分与MHC-Ⅰ类分子结合的抗原肽;外周血单个核细胞可培养出DC;效靶比40:1时负载膀胱癌抗原肽的DC诱导特异性CTLs可杀伤高达(78.5±7.0)%的BI U-87细胞。结论弱酸可以有效洗脱肿瘤细胞表面的Ⅰ类抗原肽,负载该类多肽的DC在体外可诱导出高效而特异的抗膀胱癌效应。

     

    Abstract: Objective To observe specific cytotoxicity induced by dendritic cells pulsed with acid eluted tumor antigen peptides against bladder cancer cell line.
