重组人骨形成蛋白-2(rhBMP-2)对人脑胶质瘤细胞凋亡的影响

Effects of Recombinant Human Bone Morphogenetic Protein-2(rhBMP-2) on Apoptosis of Glioma Cell SHG-44 in Vitro

  • 摘要: 目的 研究BMP-2是否对胶质瘤细胞也具有诱导凋亡作用。方法 培养的人脑胶质瘤细胞系SHG-44中加入rhBMP-2,运用透射电镜观察凋亡的超微结构,流式细胞仪技术分析细胞DNA周期变化,琼脂糖凝胶电泳检测DNA片段的改变。结果 电镜下可见细胞核染色质浓缩、边集、核碎裂;DNA周期分析显示在G1期峰前存在一个凋亡峰(12.1%);DNA电泳呈梯状。结论 rhBMP-2在体外对人脑胶质瘤细胞系SHG-44有明显诱导凋亡的作用。

     

    Abstract: Objective To investigate the effects of rhBMP-2 on apoptosis of SHG-44 glioma cell. Methods We incubated cultured human SHG-44 glioma cell with.rhBMP-2 protein.The ultrastructural changes of the SHG-44 cell were examined by electron microscopy.The cell cycle was detected by flow cytometry and changes of DNA segment were analyzed by gel electrophoresis. Results The ultrastructural changes showed compaction of nuclear chromation.An apoptosis peak in sub G1 phage was observed.The agarose gel electrophoresis of cellular DNA showed a characteristic “ladder”. Conclusion rhBMP-2 can induce apoptosis of human SHG-44 glioma cell in vitro.
