Effects of antisense oligonucleotides of BRCA1 gene on human osteosarcoma cells

  • 摘要: 目的 探索抑癌基因BRCA1基因反义寡核苷酸对人骨肉瘤细胞SOSP 96 0 7的影响。方法 通过细胞培养, 测量细胞生长曲线、计算细胞集落形成率、检测细胞周期等, 观察BRCA1基因反义寡核苷酸对人骨肉瘤细胞系SOSP 96 0 7的影响。结果 BRCA1基因反义寡核苷酸浓度为 1 0 μmol/L时, 骨肉瘤细胞的生长速度变快, 集落形成率明显增高 ;流式细胞仪检测发现其引起细胞周期变化, 细胞增殖活性提高。结论 BRCA1基因反义寡核苷酸对人骨肉瘤细胞SOSP 96 0 7有明显的影响, 使得骨肉瘤细胞恶性生物学特性更加显著, 提示BRCA1基因在人骨肉瘤发展过程中起作用

     

    Abstract: Objective To evaluate the effects of antisense oligonucleotides of BRCA1 gene in human osteosarcoma cells SOSP 9607. Methods The SOSP 9607 cell line was cultured, then the effects of the antisense oligonucleotieds of BRCA1 gene in the cell was observed by clone formatting, cell growth line, and cell cycle. Results Facilitate effects were observed by the growth line and clone formatting while it is 10μmol/L antisense oligonucleotides of BRCA1 gene.
