微卫星BAT-26 位点单态性的分析

  • 摘要: 目的 微卫星不稳定性 (Microsatellite Instability,MSI)在许多肿瘤的发生中具很重要的作用, 大多数微卫星 (Microsatellite,MS)位点在正常人群中多态性明显, 有报道表明 BAT- 2 6位点单态性很好, 据此我们分析了 60例正常肠粘膜标本中 BAT- 2 6位点 MSDNA的情况, 以便了解 BAT- 2 6位点单态性情况。方法 组织 DNA提取后 PCR扩增, 扩增产物行 6%变性聚丙烯酰胺凝胶电泳, 最后银染。结果 所有标本在 BAT- 2 6位点 MSDNA长度无明显改变。结论 BAT- 2 6具有较好的单态性。

     

    Abstract: Objective MSI plays an important role in carainogenesis of mant tumors.Most MS loci show polymorphism in normal tissues.It has been reported that BAT-26 was quasi-monomorphic,showing only minor size variathions.To analysis monomorphism at BAT-26 loci,60 normal colorectal tissues were examined. Methods Genomic DNA extracted from tissues was PCR amplified.PCR products were run on 6% denaturing polyacrylamide gel and stained with silver. Results All specimens showed no variation in the length of MS DNA at BAT...
