微缺氧对三氧化二砷抑制胃癌细胞SGC-7901生长的影响

Inhibitoy Effect of Arsenic Trioxide on Gastric Cancer Cell SGC-7901 During Hypoxia

  • 摘要: 目的 常氧和微缺氧条件下三氧化二砷(As2O3)注射液对人胃癌细胞株SGC-7901生长抑制作用的对比研究。方法 运用产气袋法(GasPaK)制造微缺氧条件,实验分为微缺氧组和对照组,用MTT法检测药物效应。流式细胞仪分析细胞周期和细胞凋亡。结果 不同浓度As2O3对人胃癌细胞株SGC-7901生长有抑制作用,并呈剂量一效应关系。细胞滞留于G2/M及S期。微缺氧条件下As2O3对人胃癌细胞株SGC-7901生长抑制作用下降(P〈0.01)。凋亡细胞百分比下降。结论 As2O3对人胃癌细胞生长有抑制作用,但对缺氧胃癌细胞的抑制作用下降,单用As2O3治疗胃癌存在一定的局限性。

     

    Abstract: Objective  To investigate the effect of arsenic t rioxide (As2O3 ) on gast ric cancer cell SGC-7901 during the normal and hypoxia. Methods  The environment of hypoxia was established by GasPak method. The effect of As2O3 was determined by MTT, and FCM was used for studying the apoptosis of SGC-7901 and cell cycle. Results  As2O3 can inhibit significantly the growth of SGC-7901, and arrest the cell in G2/M and S phases. But this inhibitory effect was decreased significantly during hypoxia ( P < 0. 01), which was found by the lower percentage of apoptosis. Conclusion  As2O3 can inhibit the growth of gastric cancer cell, but during hypoxia this inhibitory effect was decreased, so it may be limited to cure hypostatic neoplasm for only using As2O3 .
