p16 INK-4多克隆抗体的制备及在胃癌组织中的应用

摘要: 目的 制备抗 p1 6 INK- 4多克隆抗体, 并探计 p1 6蛋白的表达与胃癌预后的关系。方法 以人工合成 p1 6 INK- 4N-末端 1 3个氨基酸多肽与匙孔虫戚血蓝蛋白 (KLH)交联物为抗原, 制备抗 p1 6多克隆抗体 (Po Abs)。使用此多抗, 采用 S—P免疫组织化学方法, 检测 30例胃癌组织中 p1 6蛋白的表达。结果 30例胃癌组织中, 9例 p1 6表达阴性随访 8年死亡率 1 0 0 %、中位生存期 8个月 ;2 1例表达阳性死亡率 5 7.1 %、中位生存期 33个月, 二者生存期存在显著性差异 (P<0 .0 1 )。结论 (1 )使用自制 Po Abs可替代进口多抗体在临床、科研中的应用 ;(2 )提示 p1 6蛋白的表达与胃癌预后关系密切。Abstract: Objective To prepara Polyclonal antibodies(PoAbs) of p16 INK-4 and investigate the relationship between expression of p16 and prognosis of gastric cancer. Methods (1)PoAbs against peptide containing 13 amino acids of the N-end of human p16 was obtained in rabbits by immunization with the peptide conjugated with KLH.
