缺锌饮食对小鼠食管鳞状上皮细胞增殖的影响及机制

Effect of Dietary Zinc Deficiency on Proliferation of Esophageal Squamous Cells in Mice and Related Mechanism

    目的 探讨饮食锌缺乏对小鼠食管鳞状上皮细胞增殖的影响及其可能机制。
    方法 采用锌-小鼠-食管模型,将C57BL/6小鼠随机分为两组,每组10只,分别以足锌饲料和缺锌饲料喂养,观察小鼠体质量变化、血液和组织中的锌含量变化以及小鼠食管黏膜组织中增殖细胞核抗原(PCNA)、P38丝裂原活化蛋白激酶(P38MAPK)、核因子κB(NF-κB)p105、NF-κB p65和环氧合酶-2(COX-2)的表达水平。
    结果 与足锌组小鼠相比,缺锌组小鼠体质量、血液锌含量和食管黏膜锌含量明显下降(P < 0.05)。免疫组织化学结果显示,缺锌组小鼠食管黏膜PCNA、P38MAPK、NF-κB p105、NF-κB p65、COX-2显著增加。
    结论 饮食锌摄入不足可以抑制小鼠生长,促进小鼠食管上皮鳞状细胞增殖,其机制与诱导COX-2、P38MAPK、PCNA、NF-κBp等肿瘤相关因子过度表达有关。

     

    Abstract:
    Objective To investigate the effect of dietary zinc deficiency on the proliferation of esophageal epithelial cells in mice and related mechanism.
    Methods Twenty C57BL/6 mice were randomly divided into two groups and fed with zinc-sufficient diet and zinc-deficient diet respectively. We observed the changes of body weight, zinc concentration in blood and tissues, and the expression of PCNA, P38MAPK, NF-κB p105, NF-κB p65 and COX-2 proteins in the esophageal mucosa tissues.
    Results Compared with the zinc-sufficient group, the body weight and zinc content in blood and esophageal mucosa were significantly decreased in the zinc-deficient group (P < 0.05). Immunohistochemical results showed that PCNA, P38MAPK, NF-κB p105, NF-κB p65 and COX-2 in the esophageal mucosa of the zinc-deficient group were significantly increased.
    Conclusion Insufficient dietary zinc intake can inhibit the growth of mice and promote the proliferation of mouse esophageal epithelial squamous cells. The mechanism is related to the induced overexpression of COX-2, P38MAPK, PCNA, NF-κBp and other tumor-related factors.
