黄芪甲苷对人结肠癌SW480细胞系增殖和凋亡的影响

Regulatory Effect of Astragaloside A on Proliferation and Apoptosis of Human Colon Cancer SW480 Cell Line

    目的 探讨黄芪甲苷对人结直肠癌SW480细胞系增殖和凋亡的影响。
    方法 药物细胞毒性实验检测黄芪甲苷对SW480细胞存活率的影响,测得IC50为168 μmol/L;以不同浓度黄芪甲苷处理SW480细胞。细胞计数试剂盒检测细胞增殖倍数;蛋白质印迹法检测细胞中Ki67、PCNA、bcl-2、Bax、cleaved caspase-3和cleaved caspase-9的表达水平;Hoechst染色法检测细胞凋亡情况。
    结果 黄芪甲苷浓度大于20 μmol/L时,细胞存活率明显降低。与对照组相比,黄芪甲苷加药组细胞增殖倍数明显降低,黄芪甲苷低剂量组Ki67的表达量没有显著变化,黄芪甲苷中、高剂量组细胞中Ki67和PCNA的表达量显著降低;黄芪甲苷干预后,凋亡细胞比率显著升高,bcl-2的表达水平显著降低,而bax、cleaved caspase-3和cleaved caspase-9的表达量显著上升;且随着黄芪甲苷浓度的增加,效果越明显。
    结论 黄芪甲苷能抑制人结肠癌SW480细胞增殖,诱导细胞凋亡。

     

    Abstract:
    Objective To investigate the effect of astragaloside A on the proliferation and migration of human colon cancer SW480 cell line.
    Methods The effect of astragaloside A on the viability of SW480 cells was detected by drug cytotoxicity test. The half inhibitory concentration was 168 μmol/L. Cell proliferation rate was detected by CCK-8. The expression levels of Ki67, PCNA, bcl-2, bax, cleaved caspase-3 and cleaved caspase-9 were detected by Western blot. The apoptosis of SW480 cell was detected by Hoechst staining.
    Results The viability of SW480 cell was significantly decreased while the concentration of astragaloside was greater than 20 μmol/L. Compared with the Control group, the proliferation rate was significantly decreased after SW480 cells were treated with astragaloside A. There was no significant change in the expression level of Ki67 in the low-dose astragaloside A group. The expression levels of Ki67 and PCNA in the middle-dose and high-dose astragaloside A groups were significantly decreased. After the intervention of astragaloside A, the ratio of apoptotic cells was significantly increased, the expression level of bcl-2 was significantly decreased, the expression levels of bax, cleaved caspase-3 and cleaved caspase-9 were significantly increased, and the effect was more obvious with the increase of astragaloside A concentration.
    Conclusion Astragaloside A could inhibit the proliferation and induce the apoptosis of human colon cancer SW480 cell line.
