Correlation of FoxO3 Gene with Clinicopathological Features and Prognosis of Bladder Cancer Patients

    目的 探讨FoxO3在膀胱癌中的表达及其与患者预后和临床病理特征的相关性。
    方法 收集111例膀胱癌患者的癌组织及癌旁正常组织标本,采用实时定量PCR、免疫组织化学方法和Western blot法检测FoxO3的表达。分析FoxO3与膀胱癌临床病理特征的相关性。对FoxO3进行Kaplan-Meier法生存分析。采用Cox比例风险模型分析影响预后的独立因素。
    结果 FoxO3基因的mRNA和蛋白在膀胱癌组织中的表达均低于癌旁正常组织,差异有统计学意义(P < 0.05)。膀胱癌淋巴结转移组的FoxO3蛋白的阳性率显著低于无淋巴结转移组(P < 0.05)。TNM分期中Tis~T1浅表性膀胱癌组FoxO3蛋白的阳性率显著低于T2~T3浸润性膀胱癌组。组织学分型中高级别组的FoxO3蛋白阳性率显著低于低级别组(P < 0.05)。膀胱癌组织中FoxO3蛋白表达与疾病的恶化程度呈正相关(P < 0.05)。FoxO3蛋白表达阴性、淋巴结转移是膀胱癌患者预后不良的危险因素。
    结论 FoxO3蛋白表达在膀胱癌组织中显著低表达,其阴性表达是膀胱癌患者预后不良的危险因素。

     

    Abstract:
    Objective To investigate FoxO3 expression in bladder cancer and its correlation with clinicopathological features and prognosis.
    Methods Bladder cancer and adjacent tissue specimens were collected from 111 patients with bladder cancer. FoxO3 expression was detected by RT-qPCR, immunohistochemistry and Western blot. The relationship between FoxO3 expression and clinicopathological features and prognosis of bladder cancer patients was investigated. Kaplan-Meier survival analysis was performed on FoxO3, and Cox proportional hazards model was used to analyze the independent prognostic factors for bladder cancer.
    Results The mRNA and protein expression of FoxO3 in bladder cancer tissues was down-regulated, compared with adjacent tissues (P < 0.05). The positive rate of FoxO3 protein in patients with lymph node metastasis was significantly lower than that in patients without lymph node metastasis (P < 0.05). The positive rate of FoxO3 protein was significantly lower in patients with Tis-T1 superficial bladder cancer than that in patients with T2-T3 invasive bladder cancer; and it was also decreased in the patients at middle and high histological classification, compared with those at low histological classification (P < 0.05). The positive rate of FoxO3 was positively correlated with deterioration of bladder cancer (P < 0.05). The negative expression of FoxO3 and lymph node metastasis were considered as the risk factors for poor prognosis of bladder cancer patients.
    Conclusion Low expression of FoxO3 is identified as an important regulator in bladder cancer. Negative FoxO3 expression is risk factor for poor prognosis of patients with bladder cancer.
