烟酰胺磷酸核糖基转移酶在结肠癌中的表达、功能及生物信息学分析

Expression, Function and Bioinformatic Analysis of Nicotinamide Phosphoribosyl Transferase in Colon Cancer

    目的 探讨NAMPT在结肠癌组织中的表达并利用生物信息学分析其理化性质、蛋白结构、互作蛋白、细胞定位及对其功能进行预测分析。
    方法 利用TCGA数据库及即时聚合酶链锁反应(qPCR)技术验证NAMPT在结肠癌组织中的表达;利用siRNA敲除SW480细胞中NAMPT表达后测定细胞增殖能力;利用生物信息学ProtParam、Jpred4.0、Genecards、String等软件对NAMPT理化性质及功能进行预测分析。
    结果 在结肠癌组织中,NAMPT mRNA水平表达明显高于相应的癌旁组织;沉默SW480细胞中NAMPT后细胞增殖能力减弱;生物信息学提示NAMPT由491个氨基酸组成,为不稳定酸性亲水性蛋白质;二级结构预测发现其有14个α螺旋,17个β折叠;三级结构全球性模型质量估测(GMQE)评分0.97;预测结果显示NAMPT并无功能结构域,其仅有一个保守结构域。蛋白定位发现其在细胞外、细胞核以及胞质中均有大量表达,其参与的生物学活动主要包括参与NAD+生物合成、昼夜节律、应激、代谢反应及衰老等。
    结论 在结肠癌患者组织中NAMPT高表达,可参与多种生物学过程,并可通过不同途径调节肿瘤生物学行为。

     

    Abstract:
    Objective To investigate the expression of nicotinamide phosphoribosyl transferase(NAMPT) in colon cancer tissues and analyze its physicochemical properties, secondary and tertiary structures, protein-protein interaction, subcellular localization and function by bioinformation.
    Methods TCGA database and real-time polymerase chain reaction (qPCR) were used to verify the expression of NAMPT in colon cancer tissues. CCK8 test was used to verify the proliferation ability of SW480 cells transfected with NAMPT siRNA and control siRNA. Then, we applied bioinformatics software, such as ProtParam, Jpred4.0, Genecards, String, etc., to predict the physicochemical properties and functions of NAMPT.
    Results The expression of NAMPT mRNA was significantly higher in colon cancer tissues than that in the adjacent tissues. The proliferation ability of SW480 cells transfected with NAMPT siRNA was much lower than control group. Bioinformatics analysis indicated that NAMPT was unstable acidic hydrophilic protein and consisted of 491 amino acids. Fourteen α helixes and seventeen β folds were predicted in the secondary structure. The global model quality estimation (GMQE) score of the tertiary structure was 0.97 and the Results showed that NAMPT had no functional domain, with only one conservative domain. NAMPT was abundant in the extracellular, nucleus and cytoplasm. This protein was involved in NAD+ biosynthesis, circadian rhythm, stress, metabolism reaction and aging.
    Conclusion The overexpression of NAMPT in colon cancer tissues could be involved in various biological processes, and could regulate the biological behavior of tumor by different pathways.
