石蒜碱诱导肺癌NCI-H460细胞凋亡作用及其机制

Apoptosis Induction of Lycorine on Lung Cancer NCI-H460 Cells and Related Mechanism

    目的 通过研究石蒜碱在体外对人肺癌NCI-H460细胞增殖抑制和诱导凋亡的作用,探讨石蒜碱抗肿瘤机制。
    方法 MTT法检测石蒜碱对NCI-H460细胞增殖的抑制作用,流式细胞术检测NCI-H460细胞的凋亡率,比色法检测凋亡因子相关因子Caspase 3活性。
    结果 石蒜碱能够有效抑制肺癌NCI-H460细胞增殖,IC50为(5.79±0.11)μmol/L,对NCI-H460细胞的凋亡具有诱导效应,可增强肺癌Caspase 3的活性。
    结论 石蒜碱在体外能有效地抑制NCI-H460细胞增殖并促进其凋亡,其机制可能与激活Caspase 3的表达从而诱导肿瘤细胞凋亡有关。

     

    Abstract:
    Objective To investigate the effect of lycorine on the proliferation inhibition and apoptosis induction in human lung cancer NCI-H460 cells, and to explore the anti-tumor mechanism of lycorine.
    Methods MTT assay was used to detect the proliferation of NCI-H460 cells. The apoptosis rate of NCI-H460 cells was detected by flow cytometry. The activity of Caspase 3 was detected by colorimetry.
    Results Lycorine could inhibit the proliferation of NCI-H460 cells, and the IC50 was (5.79±0.11)μmol/L. Lycorine had an induced effect on the apoptosis of NCI-H460 cells. Lycorine could enhance Caspase 3 activity.
    Conclusion Lycorine could effectively inhibit the proliferation and induce the apoptosis of NCI-H460 cells in vitro, and its mechanism may be related to the activation of Caspase 3 expression and the apoptosis induce of tumor cells.
