基于液相色谱-质谱联用技术的男性肺腺癌患者代谢组学研究

Metabonomics Investigation of Lung Adenocarcinoma in Male Patients Based on Liquid Chromatography-mass Spectrometry

    目的 运用代谢组学方法对男性肺腺癌患者血浆代谢谱进行研究,为前期诊断提供依据。
    方法 采用基于液相色谱-质谱(LC-MS)联用技术的代谢组学方法对20例健康者和17例肺腺癌患者血中小分子代谢产物进行分析,利用主成分分析法(PCA)和偏最小二乘法(PLS-DA)进行数据模式识别,根据组间代谢群组的差异探讨肺腺癌患者的代谢特征。
    结果 多变量统计分析结果显示,健康者与肺腺癌患者样本存在明显差异,根据t检验结果在血浆中找到了16个差异代谢产物。
    结论 结合代谢通路分析,肺腺癌患者血浆中溶血磷脂酰胆碱、酯酰肉碱、脂肪酸等代谢通路出现紊乱,经鉴定的差异代谢物可作为肺腺癌早期诊断的参考。

     

    Abstract:
    Objective To investigate the plasma metabolic profiles of lung adenocarcinoma in male patients by rapid resolution liquid chromatography coupled with time-of-flight tandem mass spectrometry(RRLC-Q-TOF-MS)-based metabolomics method, to provide the basis for early diagnosis.
    Methods Metabolomics analysis was carried out for serum samples between 20 healthy controls and 17 lung adenocarcinoma patients. Principal component analysis (PCA) and partial least squares-discriminant analysis (PLS-DA) were carried out for pattern recognition and characteristic metabolites identification. Metabolic characteristics of patients with lung adenocarcinoma were investigated according to the differences of metabolic groups.
    Results Multivariate statistical analysis yielded a significant distinction in metabolic profiles between healthy and lung adenocarcinoma groups. The t-test results indicated a total of 16 biomarkers with statistically significant differences.
    Conclusion Based on the analysis of metabolic pathways, lysophosphatidylcholine, acylcarnitine, fatty acid and other substances in lung adenocarcinoma patients are disordered. These different metabolites could be helpful for diagnosing lung adenocarcinoma.
