胶原翻译后修饰酶与肿瘤关系的研究进展

Progress on Relationship Between Posttranslational Modification Enzymes of Collagen and Tumor

  • 摘要: 胶原的沉积以及硬化可以促进肿瘤的进展。胶原的生物合成包括转录、翻译和修饰,其中胶原的翻译后修饰由多种酶催化,修饰后的胶原稳定性增强、生物学功能得以保证。研究表明,胶原翻译后修饰的关键酶如脯氨酰羟化酶(prolyl-4- hydroxylase, P4H)、赖氨酰羟化酶(lysyl hydroxylase, LH)以及赖氨酰氧化酶(lysyl oxidase, LOX)与肿瘤关系密切,其异常表达能使胶原发生重构、硬化,进而促进肿瘤的发生、发展、侵袭和转移。此外,这些酶还可能是治疗癌症的潜在靶点。本文就这三种酶的基本特点及其与肿瘤进展的联系作一综述。

     

    Abstract: The deposition and stiffness of collagen promote tumor progression. As one of the processes in collagen biosynthesis, post-translation modifying of collagen is critical for collagenic stability. Three key enzymes catalyze collagen modification, such as prolyl-4-hydroxylase(P4H), lysyl hydroxylase(LH) and lysyl oxidase(LOX). Studies indicated that high levels of these enzymes could promote the initiation, the development, the invasiveness and the metastasis of tumor by inducing collagen remodeling and deposition. What’s more, it suggested that these enzymes may be potential targets for cancer therapy. In this review, we will focus on the basic characteristics of these enzymes and their associations with tumor progression.
