人端粒结合蛋白CST复合体的研究进展

Review on Human Telomere-binding Proteins of CST Components

  • 摘要: 端粒是线性染色体末端的重要保护结构,由端粒DNA和多种端粒结合蛋白构成,参与了细胞的衰老、凋亡和永生化。端粒结合蛋白在端粒沉默、端粒末端的保护及端粒长度的调节中起了重要的作用。既往研究普遍认为:酵母和脊椎动物的核心端粒结合蛋白分别为CST复合体(Cdc13、Stn1及Ten1)和shelterin复合体(TRF1、TRF2、TIN2、Rap1、TPP1和POT1)。然而,最近的研究发现CST 样复合体也同样存在于许多高等真核生物细胞中。本文概述了近年来国内外关于不同类型真核生物CST复合体结构和功能的研究进展,以揭示其在肿瘤防治中的潜在应用价值。

     

    Abstract: As an important structure contributing to linear chromosome end protection, telomere which is composed of telomeric DNA and a suite of telomere-binding proteins (TBPs) is involved in senescence, apoptosis and immortalization of cells. Telomere-binding proteins play pivotal roles in telomere silencing, telomere end protection and telomere length regulation. Previous studies have demonstrated that there are two major distinct telomere-binding complexes, CST components (Cdc13, Stn1 and Ten1) in yeast and shelterin components (TRF1, TRF2, TIN2, Rap1, TPP1 and POT1) in vertebrates. However, recent data have showed that CST-like components also exist in a wide range of higher eukaryotes. This review has summarized the research advances of construction and function of CST components in diverse eukaryotes in recent years to reveal its potential applications in tumor treatments and preventions.
