促凋亡蛋白Siva1的作用机制及其在肿瘤中的研究进展

Role of Pro-apoptotic Protein Siva1 in Tumor

  • 摘要: Siva1是凋亡诱导因子Siva编码的一种选择性剪接体,其不仅可以与多种受体如CD27、Bcl-2 或Bcl-xL、c-Abl、LPA2结合而发生相互作用,并通过调控ARF-MDM2-p53通路、抑制NF-κB的活性和酪氨酸磷酸化等方式促进细胞的凋亡,而且还可以通过抑制微管解聚蛋白Stathmin的活性和稳定微管来抑制上皮-间质细胞转化和肿瘤细胞转移。此外,Siva1可作为肿瘤患者顺铂化疗的增效剂。当今,恶性肿瘤已严重危害人类健康,其发病人数正逐年增加,故对Siva1分子机制的深入研究,必将为肿瘤的靶向治疗提供新的思路和广阔的应用前景。因此,本文就目前Siva1在国内外的相关研究进展作一综述。

     

    Abstract: Siva1 is an alternative splice form of the protein encoded by apoptosis-inducing factor Siva. Siva1 can not only bind to and interact with a variety of receptors, such as CD27, BCL-2 or Bcl-xL, c-Abl, LPA2 receptor, but also can promote cell apoptosis by regulating ARF-MDM2-p53 pathway, inhibiting the activity of NF-κB and tyrosine phosphorylation, and also can suppress epithelial-mesenchymal transition and metastasis of tumor cells by inhibiting the activity of microtubule destabilizing protein Stathmin and stabilizing microtubules. In addition, Siva1 can be used as a synergistic agent of cisplatin chemotherapy for patients with tumor. Nowadays, with the increasing morbidity, malignant tumor has seriously harmed human health. The molecular mechanism of Siva1 is very important to provide new methods and application prospects for tumor targeted therapy. Therefore, this paper reviews domestic and overseas research progresses of Siva1.
