虎纹捕鸟蛛毒素对人结肠癌SW480细胞体外增殖的影响

Effects of Ornithoctonus Huwena Venom on Proliferation of Human Colon Carcinoma Cell SW480 in vitro

  • 摘要: 探讨 虎纹捕鸟蛛毒素对结肠癌SW480细胞体外增殖的影响。方法 以体外培养的人结肠癌SW480细胞株为实验模型,以不同浓度的虎纹捕鸟蛛毒素对SW480细胞进行体外干预,通过MTT药物敏感实验检测其抑制率及计算IC50;镜下观察经Hoechst 33342染色后的细胞形态变化;流式细胞仪观测经AnnexinⅤ-FITC/PI双染的细胞状态的变化;分光光度法测定Caspase-3的相对活化倍数的变化。结果 MTT药物敏感实验显示,5~100 mg/ml虎纹捕鸟蛛粗毒对人结肠癌SW480细胞有抑制作用,且呈一定剂量效应关系。其48 h半抑制浓度(IC50)为43.68 mg/ml;显微镜下可见虎纹捕鸟蛛粗毒干预后的SW480细胞数量明显减少,细胞形态各异,并出现细胞碎片;流式细胞结果显示:10 mg/ml虎纹捕鸟蛛粗毒诱导的SW480细胞凋亡率最高,达(57.66±20.44)%;50 mg/ml虎纹捕鸟蛛粗毒干预后,SW480细胞坏死率最高,达(25.36±15.03)%;100 mg/ml虎纹捕鸟蛛粗毒干预后Caspase-3活化最明显。结论 虎纹捕鸟蛛粗毒对人结肠癌SW480细胞体外增殖具有抑制作用。

     

    Abstract: To investigate the effect of ornithoctonus huwena venom on proliferation of human colon carcinoma cell SW480 in vitro. Methods In this study, human colon carcino SW480 cell lines were cultured in vitro as the experimental model. Human colon carcino SW480 cells were intervened by different concentration solutions of ornithoctonus huwena venom in vitro. MTT chemosensitivity test was used to evaluate the suppression ratio and IC50. Cell morphology changes were observed through the microscope after dyeing by Hoechst 33342. Cells state and quantity were observed through flow cytometry after dyeing by AnnexinⅤ-FITC/PI. Caspase-3 relative activating in cell was evaluated by spectrophotometry. Results MTT chemosensitivity test results showed that human colon carcino SW480 cell quantity was reduced when intervened by 5 mg/ml-100 mg/ml of huwentoxin, with IC50=43.68 mg/ml(48 h). Microscopic observation results showed that SW480 cell quantity was reduced, with various shapes of cells and part of the cells were broken. Flow cytometry results showed that SW480 cell intervened by 10 mg/ml of huwentoxin had the highest rate of apoptosis (57.66±20.44) % and that intervened by 50 mg/ml of huwentoxin had the highest rate of Necrosis (25.36±15.03) %. Activated caspase-3 was detected out when intervened by 100 mg/ml of huwentoxin in SW480 cell. Conclusion Proliferation of human colon carcinoma cell SW480 cell can be suppressed by ornithoctonus huwena venom in vitro .
