RKIP、MMP-2与非小细胞肺癌侵袭转移的关系

Study on Expression of RKIP and MMP-2 in NSCLC Invasion and Metastasis

  • 摘要: 目的 检测非小细胞肺癌组织中Raf激酶抑制蛋白(raf kinase inhibitor protein,RKIP)和基质金属蛋白酶-2( matrix metalloproteinase-2,MMP-2)的表达及其与临床病理特征及预后的关系。方法应用免疫组织化学方法(SP法)检测83例非小细胞肺癌及其癌旁正常肺组织中RKIP与MMP-2蛋白的表达情况。结果83例非小细胞肺癌中,RKIP及MMP-2蛋白的表达率分别为20.5% (17/83)和69.9% (58/83),其表达水平与非小细胞肺癌的分化程度、TNM分期、淋巴结或远处转移及术后生存时间有明显关系;RKIP蛋白的表达水平与MMP-2呈负相关(P<0.05);RKIP蛋白低表达与MMP-2蛋白高表达的病例2年生存率明显降低(P<0.05)。结论RKIP低表达与非小细胞肺癌的侵袭、转移密切相关,RKIP可能是非小细胞肺癌的一个转移抑制蛋白,RKIP表达下调导致MMP-2的过表达从而促进非小细胞肺癌细胞的侵袭和转移。

     

    Abstract: Objective To investigate the expression of raf kinase inhibitor protein (RKIP) and matrix metalloproteinase-2(MMP-2)in non-small cell lung cancer(NSCLC) and their correlation with the clinicopathologic features and prognosis. Methods RKIP and MMP-2 protein expression of 83 cases of lung carcinoma specimens and non-tumor lung samples around were investigated by using immunohistochemistry. Results Immunohistochemistry results revealed that of the 83 cases,there was a low expression between RKIP and a high expression of MMP-2,20.5% (17/83) and 69.9% (58/83) respectively.There was correlation between RKIP and MMP-2 protein expression and differentiation of tumor,TNM staging,development of lymph node metastasis and the survival time(P<0.05,respectively ).RKIP expression was negatively correlated with MMP-2 expression in NSCLC(P<0.05);the 2-year survival rate with low level of RKIP protein was much more decreased than those with high level of MMP-2 protein(P<0.05). Conclusion The low expression of RKIP was closely correlated with invasion and metastasis of NSCLC.RKIP may be a NSCLC metastasis suppressor, whose down-regulation expression would lead to the over-expression of MMP-2,as a result,promoting the invasion and metastasis of lung carcinoma cells.
