灯盏花素对顺铂肾损害的防治及抗氧化的实验

Breviscapine Prevents and Treats Renal Injury of Cisplatin through Antioxidant in Mice

  • 摘要: 目的 观察灯盏花素对小鼠顺铂(cisplatin,DDP)肾损害的防治及抗氧化作用,探究其可能的作用机制。方法以 DDP 8 mg/kg单次腹腔注射制备小鼠肾脏损害模型,再以不同剂量的灯盏花素灌胃,1次/天,连续给药7 d后采样,观察肾脏结构变化,检测血清中肌酐(Scr)、尿素氮(BUN)、丙二醛(MDA)含量及超氧化物歧化酶(SOD)活性,肾皮质 SOD 及 MDA 的变化。 结果灯盏花素可明显改善顺铂肾损害的小鼠肾脏结构,降低血清Scr、BUN及肾皮质 MDA含量(P<0.05,P<0.01),而 SOD 活性明显升高(P<0.05)。 结论灯盏花素可明显减轻顺铂引起的肾毒性,其机制可能与抑制顺铂肾损害所致血液和肾皮质脂质过氧化反应增强有关。

     

    Abstract: Objective To observe the effects of breviscapine on cisplatin-induced nephrotoxicity in mice and possible mechanisms. Methods Renal injury were induced by a single injection of cisplatin(8mg/kg i.p),then the mice were given different doses of breviscapine (i.g) once a day for seven days..Serum BUN and Scr levels and activities of SOD and MDA in kidney cortex homogenate were determined. Results The pathological analysis indicated that renal structure was significantly improved by breviscapine.Both BUN and Scr were significantly decreased in serum(P<0.05).MDA of kidney cortex was inhibited (P<0.01),contrastly the activity of SOD increased in kidney tissue(P<0.05). Conclusion Breviscapine could reduce nephrotoxicity from cisplatin through inhibition of lipid peroxidation in the blood and renocortical tissue.
