姜黄素对乳腺癌细胞VEGF-C表达及增殖、侵袭性的影响

Effect of Curcumin on VEGF-C Expression,Proliferation and Invasion of Breast Cancer Cell

  • 摘要: 目的探讨姜黄素对乳腺癌MCF-7细胞的增殖、侵袭性及VEGF-C表达的影响。方法MTT法,Transwell小室检测姜黄素对MCF-7细胞增殖和侵袭性的影响;RT-PCR、Western blot方法检测姜黄素作用MCF-7细胞前后VEGF-C mRNA 及蛋白水平表达的变化。结果姜黄素能有效抑制乳腺癌MCF-7细胞增殖,其IC50为37μmol/L,随浓度增高其抑制作用增强(P<0.05),同时降低乳腺癌MCF-7细胞的侵袭性(P<0.001)。不同浓度姜黄素可降低MCF-7细胞VEGF-C mRNA及蛋白水平的表达(P<0.05),40 μmol/L组比20 μmol/L组下降明显(P<0.05)。结论姜黄素可以抑制乳腺癌MCF-7细胞的增殖及侵袭性,其机制可能是通过下调VEGF-C的表达。

     

    Abstract: ObjectiveTo evaluate the effects of curcumin on proliferation, invasion and VEGF-C expression in human breast cancer cell lines MCF-7. Methods Proliferation and invasion of MCF-7 cell inhibited by curcumin were assessed by MTT and Transwell technique.Expression levels of VEGF-C mRNA and protein in MCF-7 cell were determined by reverse transcription polymerase chain reaction(RT-PCR)and Western blot respectively. Results The Curcumin markedly inhibited proliferation capacity of MCF-7 cell (P<0.001).The half maximal inhibitory concentration (IC50) was 37 μmol/L.The number of invasion of MCF-7 cell were significantly lower in curcumin group than that in control group(P<0.001).It was also lower in 40 μmol/L group than 20 μmol/L group(P<0.001).VEGF-C mRNA and protein expression were also effectively down regulated in vitro.The relative expression of VEGF-C mRNA and protein were lower in 40 μmol/L group than 20 μmol/L group(P<0.05). Conclusion The proliferation and invasion ability of MCF-7 cell is effectively inhibited by curcumin.It is possible to inhibit the invasion of MCF-7 cell by down-regulation of VEGF-C expression.
