扶正解毒颗粒对染镍大鼠肾细胞DNA单链断裂的影响

Effects of Fuzheng Jiedu Granula on Nickel-induced DNA Damage in Rrats Renal Cells

  • 摘要: 目的研究扶正解毒颗粒(FJG)对镍(NiSO4)诱发大鼠肾细胞DNA损伤的拮抗作用。方法以Wistar大鼠NiSO4 [2.5 mg/(kg·d)]染毒造模,FJG灌胃处理,分别以单细胞凝胶电泳(SCGE)技术及比色法检测肾组织细胞DNA单链断裂及总抗氧化力(TAC)。结果与NiSO4组相比,FJG各组“彗星”样细胞出现率(计数一定量细胞中彗星样细胞所占的比例)、尾长、Olive尾矩、尾DNA%、尾长/头长比均明显降低(P<0.05和P<0.01),TAC则显著升高(P<0.05和P<0.01)。结论FJG对镍致细胞DNA损伤有一定的拮抗作用,其机制与抑制镍诱导氧化损伤有关。

     

    Abstract: ObjectiveTo study the effect of Fuzheng Jiedu Granula(FJG)on DNA damage in rats renal cells induced by Nickel sulfate(NiSO4). Methods Rats nephrotoxicity was induced by intraperitoneal injection of NiSO4[2.5 mg/(kg·d)] and different dosages of FJG [5,10,20 mg/(kg·d)]were administered by gavage. Cellar DNA single strand break(SSB) and total antioxidant capacity (TAC) of renal tissues were determined with single cell gel-electrophoresis (SCGE) assay and colorimetry,respetively. ResultsThe percentage of comet-like tail cells,length of tail,tail Olive moment,tail DNA percentage and ratio of tail/head were increased as well as TAC was increased significantly in FJG groups, compered with those in NiSO4 group(P<0.05, P<0.01). ConclusionBy inhibiting oxidative damage,FJG could play in antagonistic effect on DNA damage induced by nickel.
