Significance of Tiam1 and SNAI1 in EMT of Human Colorectal Carcinoma

  • 摘要: 目的探讨T淋巴瘤浸润转移因子1(T lymphoma invasion and metastasis factor 1,Tiam1)和锌指转录因子(zinc-finger transcription factor)SNAI1在结直肠癌中的表达及意义。方法免疫组织化学二步法检测137例正常结直肠组织、结直肠癌组织、淋巴结转移癌组织中Tiam1、SNAI1及E-钙黏蛋白(E-cadherin)的表达情况。结果Tiam1及SNAI1在结直肠癌组织中的表达高于正常结直肠组织(P=0.000;P=0.000),且与分化程度有关(P=0.014;P=0.014),淋巴结转移癌组织中的表达高于结直肠癌原发灶(P=0.002;P=0.001);E-cadherin在结直肠癌组织中的表达低于正常结直肠组织(P=0.000),且与分化程度有关(P=0.000),淋巴结转移癌组织中的表达低于结直肠癌原发灶(P=0.007);Tiam1与SNAI1正相关(r=0.584,P=0.000),Tiam1与E-cadherin、SNAI1与E-cadherin负相关(r=-0.324,P=0.000;r=-0.420,P=0.000)。结论Tiam1与SNAI1可能通过诱导EMT的发生而协同促进结直肠癌的浸润转移。

     

    Abstract: ObjectiveTo explore the significance of Tiam1 and SNAI1 in human colorectal carcinoma. MethodThe expressions of Tiam1, SNAI1 and E-cadherin were detected by immunohistochemistry in normal colorectal epithelium, colorectal carcinoma and lymphatic metastasis of colorectal carcinoma. ResultsThe expression rates of Tiam1 and SNAI1 in colorectal carcinoma were apparently higher than that in normal colorectal epithelium (P=0.000;P=0.000), and correlated with the degree of tumor differentiation (P=0.014;P=0.014),The expression rates of Tiam1 and SNAI1 in lymphatic metastasis were apparently higher than that in colorectal carcinoma (P=0.002;P=0.001).The expression rate of E-cadherin in colorectal carcinoma was remarkably lower than that in normal colorectal epithelium (P=0.000), and correlated with the degree of tumor differentiation (P=0.000). The expression rate of E-cadherin in lymphatic metastasis was remarkably lower than that in colorectal carcinoma (P=0.007). Tiam1 was positively correlated with SNAI1(r=0.584,P=0.000). Tiam1 and SNAI1 were negatively correlated with E-cadherin(r=-0.324,P=0.000;r=-0.420,P=0.000)). ConclusionTiam1 and SNAI1 may promote invasion and metastasis of colorectal carcinoma by inducing EMT.
