CDK4/CDK6 Inhibitor p20 and p21 Downregulate Migration and Invasion of Breast Cancer Cells

  • 摘要: 目的利用CKI肽类似物抑制CDK激酶活性,以明确CKI对人乳腺癌细胞的作用,探讨其对乳腺癌细胞体外侵袭转移的影响。方法利用作用于CDK4/CDK6的CKI肽类似物p20和p21,分别干预人乳腺癌MDA-MB-231细胞株24h。然后应用细胞与纤维连结蛋白黏附实验测定肿瘤细胞黏附率,通过单层细胞划痕实验观察细胞转染前后迁移能力的变化,Transwell侵袭实验检测细胞体外侵袭力。结果人乳腺癌MDA-MB-231细胞株经CKI肽类似物p20和p21分别干预后,细胞黏附、迁移及侵袭能力明显减弱。结论 CDK4/CDK6激酶对乳腺癌侵袭转移过程具有重要的作用。CKI肽类似物可以抑制乳腺癌MDA-MB-231细胞的黏附、侵袭和迁移。因此CKI肽类似物可能阻抑肿瘤细胞的转移,显示此类细胞穿膜肽在恶性肿瘤治疗上的潜在作用。

     

    Abstract: ObjectiveTo investigate the effectiveness of CDK4/CDK6 inhibitor p20 and p21 downregulating the migration and invasion of breast cancer cells. Methods Two CKI peptides (p20 and p21) were selected as CDK4/CDK6 kinase inhibitors.The human breast cancer cells (MDA-MB-231) were treated with CKI peptides (p20 and p21) (20μmol/L) for 24h respectively. The adhesion of MDA-MB-231 cell was measured with MTT assay. The monostratal wound healing test was used to determine the migration capacity of breast cancer cells. The changes in cell invasion were observed by the transwell invasion assay. Results After human breast cancer cells (MDA-MB-231) being treated with CKI Peptides (p20 and p21),the abilities of invasion and adhesion were decreased as compared with the control group (P<0.05).The migration capacity of breast cancer cells also significantly declined. Conclusion The CKI peptides (p20 and p21) (cell-penetrating peptides) can successfully declined the abilities of invasion and metastasis of breast cancer cells. Inhibiting the activation of CDK4/CDK6 kinase can restrain the metastasis and invasion of breast cancer cells.
