Expression and Significance of HIF-2α and Caspase-3 in Nasopharyngeal Carcinoma

  • 摘要: 目的 探讨乏氧诱导因子-2α(HIF-2α)和半胱氨酸天门冬氨酸蛋白酶-3(Caspase-3)在鼻咽癌组织中的表达及临床意义。方法 应用免疫组织化学法检测46例初诊鼻咽癌组织和20例鼻咽慢性炎组织中HIF-2α和Caspase-蛋白的表达。结果 HIF-2α蛋白在鼻咽癌组织中的表达高于鼻咽慢性炎性组织,而Caspase-3蛋白表达则相反(P<0.05);HIF-2α蛋白表达与患者性别、年龄、颈部淋巴结转移状态及临床分期无明显相关(P>0.05),与T分期的早晚相关(P<0.05)。Caspase-3蛋白的表达与患者的性别、年龄、T分期、颈部淋巴结转移状态及临床分期无明显相关(P>0.05)。鼻咽癌组织中HIF-2α和Caspase-3蛋白的阳性表达无相关性(r=-0.195,P>0.05)。结论 HIF-2α和Caspase-3蛋白的异常表达可能涉及鼻咽癌的发生和发展过程;HIF-2α蛋白的高表达可能与鼻咽癌恶性进展密切相关。

     

    Abstract: Abstract:Objective To explore the expression and clinical significance of HIF-2α and Caspase-3 in nasopharyngeal carcinoma(NPC). Methods The expression of HIF-2α and Caspase-3 protein were examined by immunochemistry in 46 samples of NPC and 20 samples of chronic nasopharynxitis. Results The expression of HIF-2α protein in NPC was higher than that in chronic nasopharynxitis while Caspase-3 protein was opposite (P<0.05). No statistical significance were observed between the expression of HIF-2α protein and patients gender,age,clinical staging and lymph node metastasis (P>0.05). There were significant statistical correlations between overexpression of HIF-2α protein and T stage. The expression of Caspase-3 was not significantly correlated with patients gender, age, T stage, clinical staging, and lymph node metastasis (P>0.05). There was no correlation between the expression of HIF-2α protein and Caspase-3 protein expression in NPC(r=-0.195,P>0.05). Conclusion The anomalous expression of HIF-2α and Caspase-3 may be associated with the carcinogenesis and development of NPC. There was significant correlation between the higher expression of HIF-2α protein and the malignant progression of NPC.
