Expression and Clinical Significance of Transketolase Genes in Hepatocellular Carcinoma

  • 摘要: 目的 探讨转酮醇酶基因家族在肝细胞肝癌(HCC)中的表达及其与临床病理的关系。方法 检测42例HCC组织及对应癌旁组织的转酮醇酶活性;用RT-PCR的方法检测转酮醇酶基因家族各成员(tkt、tktl1、tktl2)mRNA在HCC及癌旁组织中的表达。结果 HCC组织的转酮醇酶活性明显高于癌旁组织(P<0.05)。HCC组织中tktl1的阳性表达率明显高于tkt和tktl2(P<0.05),tktl1在HCC中的阳性表达率及表达量均明显高于癌旁组织(P<0.05),tktl1的表达同肿瘤分化程度、静脉癌栓、肝内有无卫星灶以及5年生存情况有关(P<0.05)。tktl2在HCC中的表达量高于癌旁组织(P<0.05),其表达与肿瘤分化程度有关(P<0.05)。结论 转酮醇酶与HCC的发生发展有关,tktl1的表达在肝癌转酮醇酶活性中起主要作用,且影响肝癌的分化、转移及患者预后;tktl2可能在肝癌分化中起协同作用。它们有望成为HCC治疗的新靶点。

     

    Abstract: Objective To investigate the expression and clinical significance of Transketolase genes in hepatocellular carcinoma(HCC).Methods Transketolase activity was assayed in 42 cases HCC and their adjacent tissues. The clinicopathological implications of transketolase genes (tkt, tktl1 and tktl2) mRNA expression detected by RT-PCR were studied.Results Transketolase activity in HCC was significantly higher than that in their adjacent tissues (P<0.05).The mRNA expression of tktl1 was significantly higher than that of tkt and tktl2 in HCC(P<0.05). Tktl1 and tktl2mRNA expression in HCC was significantly higher than that in their adjacent tissues(P<0.05).Tktl1 mRNA expression was related to tumor differentiation, portal vein cancer embolus, liver metastasis and 5-year survival (P<0.05). Tktl2 expression was related to tumor differentiation degree(P<0.05). Conclusion Transketolase was related to carcinogenesis, development and metastasis of HCC.The expression of tktl1 played an important role in the activity of Transketolase and contributed tumor differentiation, metastasis. tktl2 was a cooperation role in differentiation of HCC. They might be novel targets for therapy in HCC.
