mRNA Expression of Notch and Its Related Pathway Genes in Human Acute Lymphoblastic Leukemia Patients

  • 摘要: 目的:检测Notch信号通路相关基因的mRNA在B-ALL(急性B淋巴细胞白血病)中的表达水平,并探讨其在B-ALL中的作用。方法:建立实时定量RT-PCR方法,采用ABI PRISM 7700 PCR仪检测了16例初诊B-ALL患者骨髓(原始+幼稚淋巴细胞≥80%)和11例正常供者骨髓中Notch1、Notch2、Jagged1、Delta1和Hes1及内参GAPDH的表达水平,以检测基因=(检测基因拷贝数/GAPDH拷贝数)×106 计算检测基因的表达水平。结果:B-ALL组骨髓中 Jagged1基因的mRNA表达水平明显低于对照组,分别为(76.7±23.8)和(2 192.3±971.6)(P=0.000), Delat1基因mRNA表达水平B-ALL组明显高于对照组,分别为(60 316.6±8 713.5)和(2 189.8±802.3)(P=0.002), 差异有统计学意义。Notch1、Notch2和Hes1基因的mRNA表达水平B-ALL组分别为(6 167.6±1661.3)、(95 318.6±9 437.5)和(258.2±96.2),对照组分别为(1 735.5±936.6)、(52 078.2±8 746.3)和(1 006.2±806.0)(P值分别为0.089、0.815和0.786),此三个基因在B-ALL组与对照组之间表达差异没有统计学意义。结论:Jagged1在B-ALL中低表达,Delta1在B-ALL中高表达,提示B-ALL发病与Notch信号途径中配基的异常表达有关。

     

    Abstract: Objective:To investigate the possible role of Notch signaling pathway in the development of human acute B cell lymphoblastic leukemia. Methods:Real-time quantitative RT-PCR was applied to detect the mRNA levels of Notch signaling genes including Notch1,Notch2,Jagged1,Delta1,Hes1 with GAPDH as the internal control, in the bone marrow from 16 B-ALL patients at diagnose(the blast plus prolymphocytes over 80%)and 11 normal controls by using a ABI PRISM 7700 thermal amplifier. The mRNA expression level of Notch was normalized as a ratio between the PCR values of Notch and GAPDH times 106. Results:The mean value of mRNA levels of Jagged1in 16 B-ALL at diagnose was significantly lower than those of control group[(76.7±23.8) vs. (2192.3±971.6), P=0.000], while the mean value of mRNA level of Delta1in 16 B-ALL at diagnose was statistically higher than those of control group [(60316.6±8713.5) vs. (2189.8±802.3), P=0.002]. However, The mean mRNA expression levels of Notch1,Notch2 and Hes1in B-ALL group at diagnose [(6167.6±1661.3), (95318.6±9437.5) and (258.2±96.2) respectively] compared with those in normal control group [(1735.5±936.6), (52078.2±8746.3) and (1006.2±806.0) respectively]showed no statistic difference between these two groups (P= 0.089, 0.815 and 0.786 respectively). Conclusion:The mRNA level of Jagged1was dramatically less while Delta1 was evidently higher in B-ALL patients compared with those in normal control group, suggesting the aberrant expression of Jagged1 and Delta1in Notch signaling pathway may contribute to the development of human acute lymphoblastic leukemia.
