Expression of EphA1 Gene in colorectal Cancers and Its Clinical Significance

  • 摘要: 目的:探讨EphA1在结直肠癌中的表达及其临床意义。方法:利用荧光实时定量RT-PCR检测EphA1 在肠癌细胞系和组织中的表达,并分析甲基化状态。免疫组织化学染色分析EphA1蛋白在肠癌组织中的表达及其临床意义。结果:EphA1在肠癌细胞中不同程度表达,并存在甲基化。EphA1 mRNA在癌旁正常黏膜和肿瘤组织中表达差异无统计学意义(P=0.496)。EphA1蛋白在肠腺瘤中弥漫强阳性,在癌组织中主要为低或缺失表达,低表达多见于低分化腺癌(P=0.027) 、伴有更深的肠壁浸润深度(P=0.002)和淋巴结转移患者(P=0.019)。EphA1下调多见于中晚期病例(P=0.002),下调较上调患者预后不良。结论:EphA1在结直肠癌的发生发展中可能具有一定肿瘤抑制作用,有可能成为预测结直肠癌恶性程度和预后的新指标。

     

    Abstract: Objective:To investigate the expression of EphA1 in colorectal cancers and its clinical significance. Methods :Quantitative real-time RT-PCR and immunohistostaining were employed to detect the expression level of EphA1 in colon cancer cell lines and CRC specimens.Methylation specific PCR (MSP) was also performed. Results :The expression of EphA1 transcript, methylated DNA of EphA1 was detected in cell lines.EphA1 mRNA expression in the corresponding normal mucosae is not significantly different from that in tumor tissues (P=0.496).In adenomas, the immunostaining for EphA1 was positive diffusely and strongly.However, in CRCs, the expression of EphA1 protein was predominately down-regulated.Reduced or lost expression of the EphA1 protein was significantly associated with poor differentiation (P=0.027), greater depth of wall invasion (P=0.002) and advanced TNM stage (P=0.002), and preferentially occurred in patients with lymph node metastasis (P=0.019).Patients with reduced EphA1 expression had a tendency of poorer prognosis than those with EphA1 upregulation.ConcolusionEphA1 may have tumor suppressor roles in CRCs and may be an important biomarker by which malignancy, biological behavior and prognosis are evaluated.
