Expression of CD151 and Integrin α3β1 in Breast Cancer and Their Significance

  • 摘要: 目的:探讨CD151和整合素α3β1蛋白在乳腺癌发生发展和浸润转移中的作用。方法:构建包含40例正常乳腺和130例乳腺癌组织的组织芯片,免疫组织化学SP法检测CD151和整合素α3β1的蛋白表达,对临床病理资料进行比较,并分析两者的相关性。结果:CD151和整合素α3β1在正常乳腺组织中的阳性表达率分别为0(0/39)和7.7%(3/39);在原位癌组织中的阳性表达率分别为33.3%(10/30)和30.0%(9/30),在浸润性乳腺癌组织中的阳性表达率分别为59.0%(59/100)和70.0%(70/100),两种蛋白的阳性表达率在三组间比较差异有统计学意义(χ2 =41.28和χ2 =48.62)。二者的表达与乳腺癌患者的年龄、肿瘤大小和组织学类型无相关性(P>0.05); 但与淋巴结转移,肿瘤分化程度和临床分期密切相关(P<0.05),此外,CD151与ER、PR的表达有关(P<0.05)。CD151和α3β1蛋白在乳腺癌组织中的表达呈正相关(r=0.565;P<0.01)。结论:CD151和整合素α3β1在乳腺癌组织中存在异常高表达,并与乳腺癌发生发展、浸润转移密切相关。联合检测可作为判断乳腺癌生物学行为和预后的重要指标。

     

    Abstract: Objective:To investigate the role of CD151 and intergrin α3β1 proteins in the occurrence development and invasion and metastasis of breast cancer. Methods :Tissue microarray (TMA) which contained 40 cases of normal tissues and 130 cases of various kind of breast cancer was manufactured.Protein levels of CD151 and intergrinα3β1 were detected by using immunobiochemical steptavidin peroxidase method (SP).The χ2 test and Spearman analysis were used.Statistical significance was defined as a P value< 0.05. Results :The positive rates of CD151 and integrinα 3β1 expressions were 0.0%(0/39)and 7.7%(3/39) in normal breast tissues; 33.3%(10/30)and 30%(9/30) in tissues of carcinoma in situ, respectively; 59.0%(59/100) and 70.0%(70/100)in cancerous tissues, respectively.The differences between two kinds of protein expression rates among the three groups were significant (χ2 =41.28, χ2=48.62).The expressions of CD151 and intergrinα3β1 protein in breast cancers have no relationship with patient's age the histological type and size of cancer but significantly related with differentiated degree of tumor lymph nodes metastasis and clinical stage.(P<0.05).In particular, it is worth noting that there is significantly relation between the expression of CD151and ER/ PR (P<0.05).There is positive correlation between the expression of CD151 and intergrinα3β1 protein(r=0.565; P<0.01). Conclusion :The CD151 and intergrinα3β1 were over expressed in breast carcinoma, which suggested that two proteins were related to oncogenesis, development, invasion and metastasis of breast carcinoma.Joint detection of the two makers can be used to determine the biological behavior and prognosis of breast cancer.
